It depends somewhat on the sentence/paragraph it's attached to; but practically always as some form of "just kidding", somewhere in between ;) and  ;D
I think :D is just creepy looking, I don't think I've seen anyone use it here..

:P is a bit goofy looking, so it seems fitting for goofy/'tongue in cheek' remarks.

Smileys were supposed to improve the quality of communication by giving the tone of the message.  It would help to distinguish between a serious flame from a joke, or irony.  But if we don't agree on the meaning of individual smileys, there will be other misunderstandings, for instance if irony is mistaken for happiness.

For me:
;)  - "You see what I mean."  (maybe refers to some other recent topic)
:P - Something like "Admit it, I won, you lost.", or "Na na na!"

In fact it is not easy to describe the exact feeling a smiley evokes to you.
